Immortalization Method	Serial passaging and infection by SV-40 tsA209 Temperature Sensitive Mutant virus
Description	The corpus luteum develops from an ovarian follicle during the luteal phase of the menstrual cycle and is responsible for the decidualization of the endometrium. The Immortalized Rat Luteal Cells (GG-CL) is conditionally immortalized through the temperature-sensitive simian virus (SV40 tsA209) and has retained many primary luteal cell markers such as estrogen receptor-β, glucocorticoid receptor (GR), interleukin-6 (IL-6), orphan nuclear receptor nur 77 and 20α-hydroxysteroid dehydrogenase (20α-HSD). In addition, GG-CL responds to stimulation by PRL, estradiol, and cAMP, making the cells a useful model for studies involving gene expression and regulation of luteal cells.

Propagation	Use of PriCoat? T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow II medium available from ABM (TM002). To make the complete growth medium, add the following components to the base medium: 10% fetal bovine serum (TM999) and Penicillin/streptomycin. Atmosphere: air: 95%, CO2: 5%; Temperature: 33.0°C. The functional expression of the SV40 antigen is induced by culturing the cells in vitro at a temperature permissive (33°C). At a non-permissive temperature (37°C-39°C), the cells cease to proliferate and revert to differentiated phenotype.
Preservation	1. Freeze Medium: Complete growth medium with 20% FBS and 10% DMSO. 2. Storage Temperature: Liquid nitrogen vapour phase.
Quality Control	1) Northern analysis and RT-PCR were used to confirm the expression of IL-6, glucocorticoid receptor (GR), estrogen receptor-β (ER-β) and orphan nuclear receptor nur 77. 2) Cells were subjected to treatments of exogenous prolactin, estradiol, and cAMP to examine effects.
